啟動(dòng)子篇:芽孢桿菌啟動(dòng)子

代謝工程可以通過改變遺傳調(diào)控元器件的表達(dá)強(qiáng)度達(dá)到優(yōu)化代謝途徑的目的,進(jìn)而實(shí)現(xiàn)目的產(chǎn)物的高效合成。遺傳調(diào)控元器件主要包括啟動(dòng)子、核糖體結(jié)合位點(diǎn)、終止子以及其他一些調(diào)控基因表達(dá)的元件。

啟動(dòng)子的定義:

啟動(dòng)子是指一段能使基因進(jìn)行轉(zhuǎn)錄的DNA 序列,包括核心啟動(dòng)子區(qū) (?35 區(qū)、?10 區(qū)) 和調(diào)控區(qū)域。在轉(zhuǎn)錄起始時(shí),啟動(dòng)子與RNA 聚合酶、調(diào)控基因轉(zhuǎn)錄的轉(zhuǎn)錄因子產(chǎn)生相互作用, 進(jìn)而控制基因轉(zhuǎn)錄的起始時(shí)間和程度[1]。

啟動(dòng)子結(jié)構(gòu)

啟動(dòng)子的類型:

按照其誘導(dǎo)的方式可以分為組成型和誘導(dǎo)型啟動(dòng)子。
組成型啟動(dòng)子(Constitutive promoter)具有強(qiáng)啟動(dòng)、不需添加誘導(dǎo)劑、可直接產(chǎn)生目的蛋白以及成本低等優(yōu)勢(shì),常見的如P43、PHpaII、Plaps及PamyQ;根據(jù)表達(dá)時(shí)期的不同,又可分為指數(shù)階段、對(duì)數(shù)中期和早期平穩(wěn)階段、滯后對(duì)數(shù)和平穩(wěn)階段以及平穩(wěn)階段4 種類型。
誘導(dǎo)型啟動(dòng)子(Inducible promoter)可在特定的時(shí)期調(diào)控異源蛋白的表達(dá)量,具有可控、快速、大量等特點(diǎn),如Pgrac、Pspac、Pxyl等[2]。

其他誘導(dǎo)型啟動(dòng)子

  1. 蔗糖啟動(dòng)子PsacB:?jiǎn)?dòng)子的轉(zhuǎn)錄不受蔗糖的嚴(yán)格調(diào)控,在沒有誘導(dǎo)劑時(shí)也能進(jìn)行轉(zhuǎn)錄,但較有蔗糖誘導(dǎo)時(shí)強(qiáng)度低100倍。
  2. 麥芽糖誘導(dǎo)型啟動(dòng)子Pglv:glv啟動(dòng)子以麥芽糖為誘導(dǎo)物,價(jià)格便宜,工業(yè)成本低,安全無毒,使用價(jià)值比較大。但是發(fā)酵中產(chǎn)生的葡萄糖會(huì)對(duì)啟動(dòng)子有強(qiáng)烈的反饋抑制,這是工業(yè)應(yīng)用上面急需解決的一個(gè)問題。
